SELECTION OF SOIL BACTERIA FOR CONTROLLING CASSAVA MEALYBUGS

Soil bacteria are the key living organisms in several kinds of biogeochemical processing due to their production of potential hydrolytic enzymes, which have been attractive for several applications including bio-pesticides. In this study, bacteria capable of producing chitinase, proteinase, and lipase, were enumerated and isolated from soil collected from different cassava plantation areas, to obtain the dominant bacteria having small cells suitable for invading some pores of the cassava mealybugs collected from the same locations as the soil. Seven Gram-negative, non-spore forming bacterial isolates capable of producing potential hydrolytic enzymes and with small cell sizes (0.2-1.0×1.0-3.0 μm) occurring singly or in pairs, were selected. These isolates could still not be identified into species by their morphological, cultural, and biochemical characteristics, as well as 16S rRNA gene sequences. When compared to their closest relatives, the 7 isolates designated as SUTT 15 and SUTT 16, SUTT 26 and SUTS 80, SUTS 53 and SUTS 65, and SUTS 59, belonged to genera Chromobacterium, Chryseobacterium, Pseudomonas, and Serratia, respectively. Cassava mealybugs, particularly the nymph and adult of the pink mealybug (Phenacoccus manihoti Matile-Ferrero), as the bacterium-invading targets were mainly found on cassava plants and accumulated in the soil under the plants to be the nutrients.
