APOPTOSIS OF MCF-7 CANCER CELL INDUCED BY POMEGRANATE (Punica granatum L.) PEEL EXTRACT

Various aspects of pomegranate (Punica granatum Linn.) have been documented intraditional medicine. In the present study, pomegranate peel ethanolic extract (PPE) wasinvestigated for many aspects relating to the caspase-3-deficient human breast cancer cellline (MCF-7), especially on the antiproliferative effects, morphological changes, DNAfragmentation, and apoptotic molecule-activated cell death. The results showed that PPEcould inhibit the proliferation of MCF-7 cells in a dose- and time-dependent manner. PPEhas a strongly positive influence on antiproliferation and apoptotic induction of MCF-7cells, as seen via morphological changes and oligonucleosomal DNA fragments. Apoptosiswas accompanied by bcl-2, caspase-9, caspase-7, and poly (ADP-ribose) polymerase(PARP) levels. PPE induced apoptosis indicating that there was a reduction of bcl-2,procaspase-9, and procaspase-7 activities, but it activated the PARP activity. Moreover,these protein levels were associated with morphological change and DNA fragmentation inthe MCF-7 cells. However, caspase-3 deficiency did not affect the PPE-induced levels ofbcl-2, caspase-9, caspase-7, and PARP. In conclusion, the PPE had the potential for theinduction of antiproliferation and apoptosis in caspase-3-deficient MCF-7 cells.
